The principal overland route begins in Cusco and travels through Huarcapay, Ninamarca and Paucartambo before reaching Acjanaco and the cloud forest. It then descends through Pilcopata toward Atalaya on the Alto Madre de Dios River. The changing altitude is a defining part of the journey, connecting Andean landscapes with humid forest and Amazon lowlands.
For deeper Manu River itineraries, travel continues from Atalaya by motorized boat toward the park's visitor sectors. SERNANP notes that access combines road transport, river navigation and walking, so journey times depend on the selected route and current conditions.
